When an App updates through the App Store, do I have to click on the OPEN Button in order for it to take effect?

No you don't need to. The next time you open the App itself the update is there. In addition to the OPEN option in the App Store, you will also see a little green dot on the App icon of any app that has been updated but not opened yet.
