lock iPad before iCloud backup?

What does it mean to be "locked"in this context, and how do I make sure my iPad is ready to backup?

If you have iCloud Backup turned on, it will backup automatically while charging if it is locked and connect to wi-fi. If you don't have Settings > Display & Brightness > Auto-Lock set to Never, it will lock on its own.
