Track Automation or Region Automation in Mix?

Hi,

I am new in Mixing business.

What is the right way to use Automation in Track and Region?

Should be all tracks Audio or can be a combination with Audio and MIDI?

Using both types of Automation together creates contradiction and level button jumps up/down.
