Mac,Late,2009,Microphone,not,working,,Preferenc...

Microphone is not working on my iMac 21.5 Late 2009 OSX Sierra 10.12.6, RAM 4GB, 3.06 GHz, HD 500 GB 90% free storage. I checked with Preferences/Sound/Input and still not working. When I tried to start my iMac with cmd+alt+P+R, I got a white screen without logo. It is s/w problem because when a technician started my iMac from another hard disc the microphone was working. When I returned home the problem appeared again. Probably a virus. What should I do?
